Principle of the Assay::The Human DKK3 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human DKK3 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human DKK3 coated on a 96-well plate. Standards and samples are pipetted into the wells and DKK3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human DKK3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of DKK3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Dickkopf-related protein 3 is a protein that in humans is encoded by the DKK3 gene. This gene encodes a protein that is a member of the dickkopf family. It is mapped to 11p15.3. The secreted protein contains two cysteine rich regions and is involved in embryonic development through its interactions with the Wnt signaling pathway. The expression of this gene is decreased in a variety of cancer cell lines and it may function as a tumor suppressor gene. Members of the Dkk-related family display unique patterns of mRNA expression in human and mouse tissues, and are secreted when expressed in 293T cells. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation.
